Fully Validated FT-NIR Solutions

Bruker’s FT-NIR spectrometers are equipped with an automated filter wheel which houses standard materials and filters for testing instrument performance. Included in the OPUS software is OVP (Optics Validation Program), an instrument test program which executes a series of performance tests using the standards in the filter wheel. This program evaluates the instrument performance and determines if the spectrometer is operating within specifications.

OPUS fully supports the demands of the 21 CFR Part 11 regulation (Electronic Records, Electronic Signatures) issued by the FDA. Extensive user management with multiple security levels, non-editable data files and complete audit trails are some of the many features of this comprehensive spectroscopy software.

Online Monitoring of Fermentation processes

In the pharmaceutical environment, due to the strict regulatory requirements, the continuous control of fermentation processes is an essential part of the manufacturing process. For an optimal result, it is necessary to measure, control and if possible adjust several variables, like glucose, lactate, protein concentrations and more. Fermentations are initially transparent but become increasingly opaque as the cultivation progresses. Therefore, transmission measurements are unsuitable for monitoring the entire cultivation process.

To provide reproducible measurements even for slurries and emulsion, Bruker developed an outstandingly performing transflectance probe. In combination with the powerful spectrometers by Bruker, also the presence of gas bubbles will not influence the results.

Online analysis of tablets including content uniformity

Traditionally content uniformity analysis is performed by HPLC. This time-consuming, labor-intensive process requires a test samples being taken from the production area to the QC/QA laboratory where they are prepared and analyzed.

FT-NIR spectroscopy has proven to be a rapid, cost and time-effective and non-destructive alternative for routine content uniformity analysis. Bruker has developed a technical solution which combines a MATRIX-F and online content uniformity measurements with conventional tablet testing.

The TANDEM is a fully automatic on-line PAT tool that allows the collection of process data and control the tablet compression process. The system provides both physical and chemical characteristics of pharmaceutical tablets. It can be offered in two different configurations:

Stand-alone mode

  • not directly connected to a tablet press
  • manual feed of the tablets into the system
  • status and results are available as OPC outputs only
  • no control by the tablet press DCS
  • controlled by operators
  • detailed reports are available

Integrated, automatic mode

  • connected to a tablet press
  • automatic feed of the tablets into the system
  • interfaced to a tablet press via OPC (input/output)
  • controlled by tablet press DCS
  • no reports are available (single source of information is tablet press data base)

Filter Dryer Endpoint Analysis

The drying step is critical to ensure that the correct characteristics (moisture/solvent content) are controlled for the next step in the process. The standard operating methods to determine the moisture content, involves either stopping the process and thieving a sample for primary analysis or drying the product for a fixed period. Both do not provide a continuous monitor of the drying process and therefore do not provide the control necessary to optimize the product moisture.

FT-NIR spectroscopy in combination with an online diffuse reflectance probe enables a continuous monitoring the drying process. Filter driers can be modified to install measurement ports in the filter drier bowl into which the probes can be mounted. Depending on the amount of sample, the probe position can be adjusted.
